1. There are three types of API test kits discussed in today’s lab. Answer the following prompts based on these tests: Describe the type of bacteria that can be used with each test. 3. The Methyl Red and Voges Proskauer tests are conducted simultaneously at 24 hours and repeated at 48 hours after incubation. Why is this done? If you see a change in the results of your Methyl Red test at 24 hours and 48 hours, what has occurred? Which result is more accurate and why? (1 point)
4. What is the purpose of conducting the Indole test? Please describe using your own words what a positive and negative test look like and what reactions are indicated by each result. (1 point)
5. The Methyl Red test contains glucose, what other tests contain this compound? Are they comparable? Why or why not? In the MR test, some bacteria convert the glucose to many acid end products, what are these bacteria known as? (2 points)
6. Analyze your MR-VP results shown by your unknown bacteria isolate. What do each of these results indicate about the chemical pathways utilized by your bacteria? Explain. (1 point)
7. Using Bergey’s Manual, you determined that the identification for a bacterial unknown isolate was Acinetobacter baumannii. You decide to compare this result to an identification from an API test strip. You obtain the following results from an API20E test strip after incubation and the addition of all post-incubation reagents. Use the instructions manual from Canvas to fill out the API20E Blank Reading Sheet (also on Canvas) and demonstrate the results for each test and the resulting 7-digit code. (1 point)
8. Compare the results of the tests within your API test strip that were similar to biochemical tests performed throughout the semester. Analyze if each of the results match and what it says about your bacteria metabolism. (2 points)
9. You inoculate and incubate a Citrate test and receive the following result but you can’t see visible bacterial growth within the slant. What does this positive result indicate about your bacterial unknown? Is this a valid test? (1 point)
10. Analyze the results obtained for each API test within your strip (Was it a positive or negative? What does this indicate about your bacteria?). What is the final 7-digit profile you obtained for your bacteria? What identification did you receive from this 7-digit profile? How does this identification compare to what you know about your bacteria’s colony and cellular morphology? Does it match? Why or why not? (4 points)
